@mappingtoolsMapPoint lets you set the start time for the route. For multi-day routes, you can also set the daily driving time by specifying the time to stop driving. Periodic rest stops can also be specified.
All of these settings are on the Profile tab of the More route options dialog box (displayed by pressing the More options button on the Route Planner pane):
The driving times_option
Use the spin buttons to set the start and end times. The default is for a total of eight hours driving per day, but you can expand or contract this if you wish. These times are not necessarily fixed. Use the Flexibility drop-down box to change how much MapPoint is allowed to vary these times from day to day. The default is for a quarter of an hour, but the flexibility can vary from one minute to two hours. The times can also be fixed.
MapPoint can also insert periodic rest stops in the directions and route timings. These are controlled by the Automatic rest stops settings on the above dialog box. Set the check box, and use the spin buttons to set the length of each rest stop (in minutes) and the period between each rest stop (in hours and minutes).
Here are the results of a four hour interval. A 15 minute rest stop has been inserted at midday and appears on the map as a small dot:

Individual Waypoint Times
As well as setting periodic rest stops and daily start/end times, you can set the times for individual waypoints. Do this by selecting a waypoint in the Route Planner pane and pressing the ‘clock’ button (top button on the right of this pane). This will display the following Schedule Stop dialog box:

Use this dialog box to set how long a waypoint should last for, and the waypoint’s arrival and departure times. The defaults are Automatic – ie. let MapPoint choose when the route will arrive or depart the waypoint. Note that two (or more) waypoints with fixed times will usually conflict. Therefore expect MapPoint to match one exactly, and then to either insert a long wait time or even to ignore the second waypoint’s arrival time.
